Quick-Neuron™ GABAergic Neuronの免疫染色

Quick-Neuron™ GABAergic -mRNAキットで誘導開始後11日目の細胞は、神経細胞マーカーTUBB3およびGABA作動性神経細胞特異的マーカーGAD65/67を発現し、典型的な神経突起の伸長を示します(スケールバー=100μm)。

染色条件:Anti-β-III tubulin monoclonal antibody (R&D Sys- tems, Catalog Number: MAB1195, 1:250 希釈)、二次抗体(Invitrogen,Catalog Number: A32723 Goat anti-Mouse IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, AlexaFluor Plus 488, 1:500 希釈)

Anti-GAD65/GAD67 primary antibody (Fisher Scientific, Catalog Number: PA5-36080, 1:400 希釈)、二次抗体(Invitrogen, Catalog Number: A11037 Goat anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Sec- ondary Antibody, Alexa Fluor 594, 1:500 希釈)

核はHoechst33342で対比染色を行いました。

Quick-Neuron™ GABAergic - mRNA Kitsでは、お手持ちのiPS/ES細胞株を高速、簡単、かつ効率的にGABA作動性神経細胞に分化させることができます。 このキットでは、分化を誘導するための弊社独自の転写因子カクテルを合成mRNA(syn-mRNA)で導入するため、フットプリントフリーで、つまりゲノムに影響を与えずに目的の細胞へと分化させることが可能です。

CIRMのiPS細胞バンクには1,500以上のiPS細胞株が登録されていますので、特定のドナー細胞株由来のコリン作動性神経細胞をご希望の場合はお問い合わせください。当社が提供するコリン作動性神経細胞はすべて当社独自の転写因子ベースの技術を用いて分化させており、「フットプリントフリー」で機能的かつ高品質な細胞です。

Quick-Neuron™ differentiation kits にはどのようなサイズがありますか?

SmallサイズとLargeサイズをご用意しております。Smallサイズは24ウェルプレートの4ウェル分に使用できる試薬セットで、シングルセル解析、イメージング解析、パイロットスタディなどに最適です。

Largeサイズは6ウェルプレート6ウェル分または10cmディッシュ1枚分に使用できる試薬セットで、ハイスループットスクリーニングやRNAシーケンシングなどのアプリケーションなどに最適です。
